چکیده

446 Virulence Volume 3 Issue 5 Recent studies have identified cytoskeletal elements in bacteria which play important roles in cellular morphology, cell division, DNA segregation and the establishment of cell polarity. However, our understanding of the contribution the bacterial cytoskeleton makes toward virulence is lacking. The MreB protein is a bacterial homolog of eukaryotic actin and interacts intimately with MreC and MreD. We provide evidence that the Mrebased cytoskeleton directly influences pathogenicity in Salmonella. Disruption of MreC and MreD caused the downregulation of the Salmonella pathogenicity island 1 (SPI-1) type 3 secretion system (T3SS) essential for host cell-invasion, and also flagella-mediated motility. These phenotypic effects were mediated by RcsC, the sensor kinase of the Rcs phosphorelay two-component system and a major envelope stress regulator. Curiously, the SPI-2 T3SS remained viable. Our results demonstrate the importance of the integrity of the bacterial cytoskeleton for virulence, highlighting the impact of associated global regulatory mechanisms on pathogenicity. Structural homologs of all three major eukaryotic cytoskeletal protein classes are now known to exist in bacteria. These homologs include MreB and ParM (actin), crescentin (intermediate filaments) and FtsZ (microtubules). Ongoing research seeks to understand the mechanisms by which the bacterial cytoskeleton and associated proteins control cell shape, through their control of synthesis of the stress-bearing peptidoglycan cell wall (Shih and Rothfield, Microbiol Mol Biol Rev 2006). To date six essential cell shape-determinant proteins have been identified in non-spherical bacteria: MreB, MreC, MreD, penicillin binding protein 2 (PBP2), RodA and RodZ. The chemical or genetic inactivation of any of these proteins causes a loss of rod-shape and, in the absence of compensatory mutations, eventual celllysis. The cell shape-determinant proteins are thought to form a large multi-protein complex together with the peptidoglycan synthetic Virulence News & Views

Novel Inhibitors of Staphylococcus aureus Virulence Gene Expression and Biofilm Formation

Staphylococcus aureus is a major human pathogen and one of the more prominent pathogens causing biofilm related infections in clinic. Antibiotic resistance in S. aureus such as methicillin resistance is approaching an epidemic level. Antibiotic resistance is widespread among major human pathogens and poses a serious problem for public health. Conventional antibiotics are either bacteriostatic o...

RNAi technology: A Novel approaches against fungal infections

Despite the introduction of new antifungal agents, resistances to antifungal therapy continue to increase and outcome of invasive fungal infections treatment is frequently suboptimal. A large amount of the recent effort in antifungal drug discovery has focused on a limited set of targets with functions known or expected to be important for fungal viability and virulence. A variety of techniques...

Novel Oxadiazole Thioglycosides as Potential Anti- Acinetobacter Agents

The glycosylation of 1,3,4-oxadiazole-2-thiones has been performed with peracetylated β-pyranosyl bromide in the presence of potassium carbonate. Deprotection of acetylated thioglycosides was necessary for increasing their antibacterial effects. The structure of nucleosides were confirmed by 1H NMR, 13C NMR and HRMS. The anomeric protons of nucleosides c1–4 were assigned to the doublet, confirm...

Protective Effect of Hyaluronidase and Type-specific Anti-m Serum on Experimental Group a Streptococcus Infections in Mice

Five strains of encapsulated group A streptococci of different serological types, each with a glossy and a matt variant, were studied to compare the rôles of the M substance and the hyaluronic acid capsule in virulence of these microorganisms. The results indicated that both contribute to the virulence of group A streptococci but that the M antigen is the more fundamental factor. Encapsulated v...

Novel Approach Synthesis, Molecular Docking and Cytotoxic Activity Evaluation of N-phenyl-2,2-dichloroacetamide Derivatives as Anticancer Agents

Dichloroacetate (DCA) as a small, cheap and available anticancer agent, is a pyruvate mimetic compound that stimulates the activity of pyruvate dehydrogenase (PDH) enzyme through inhibition of pyruvate dehydrogenase kinases (PDHK1-4). DCA turns on programed cell death (apoptosis) which suppressed in tumor cells and therefore inhibits tumor growth. DCA also interferes with the glucose uses of ca...
